Is it possible to install ILANCE locally for testing and adapting before going online?

Yes. You can have a development instance going anywhere so long as it does not go public. No problem :)

Sure.
Basically, let's say you buy ILance Starter 3.1.5. That means you have a license to launch ILance on any server, on any domain name (and supports unlimited sub-domains) for that same license and files. In other words, you are able to install and launch ILance on unlimited number of sub-domains based on the main primary license for that domain.
Ie:
You buy a license for domain.com. Then you launch service.domain.com, product.domain.com, etc. You can have unlimited installs of ILance on any sub-domains under that main domain. That is clarification #1.
For the second, it makes perfect sense that you will not want to launch ILance on your primary production server before you even design it, modify it (if you feel the need to do so) and of course tinker here and there. It makes sense that you'll want to install some local server environment like we do.. WAMP (windows, apache, mysql and PHP) and develop, test before it reaches domain.com production.
In that case, you can do so. No problem. If you don't have access to install a local server like with WAMP and you do all your testing on your production server (maybe in a folder called /beta/ or /demo/ or /ilance/ or /test/) you can also do so. In this situation, seeing how google has magic eyes and yahoo the same, you will be required to password protect this folder where you'll be doing your development for your license.
This avoids you having a production server online with ILance, and then another instance of ILance in a folder off the production server.... that would mean you have 2 instances of ILance installed based on the Starter License you purchased. For that situation you need to deny access to anyone looking at your development area if it's sitting on your production web server.

is it necessary to duplicate database ? (for first instance dev // production)

You should have the same database which you have on the production server. Only difference should be emails. You have 2 options. Disable email sending on your localhost machine or change all emails in ilance_user table to somehing like "test". Otherwise your customers will get emails from you dev machine.

You don't need additional license for dev environment.

Q1 My subdomain need a htaccess file what modifications I must done in that file or in /function/config.php to deploy the dev.mysite.com operationnal ??
